Biochemistry, Molecular Biology & Structural Biology:
Analysis of the DNA Replication machinery
MRC Band 4 Salary £29,663 – £35,399 per annum (including location allowances)

The Clinical Sciences Centre, which is directly funded by the Medical Research Council, is a division of Imperial College. The CSC and Imperial College provide excellent core facilities and support structures. The DNA Replication Laboratory is part of the Epigenetics, Development and Cancer Section and offers interactions with groups studying cell cycle, cancer, nuclear organisation and chromatin.

We are looking to recruit a Postdoctoral Research position within the DNA Replication Group at the MRC-CSC. We are looking for a very motivated Biochemist that is interested in joining our friendly and international team. You will join a rapidly developing research project focussed on the eukarotic MCM2-7 helicase. We have previously characterised the assembly process of the ORC-Cdc6 complex on DNA (Speck et al. 2005 Nature Structural & Molecular Biology; Speck and Stillman 2007 JBC; Speck et al 2008 PNAS).

Recently, we purified the MCM2-7 helicase and showed that MCM is active in pre-Replication Complex formation. Now we aim to characterise its helicase activity alone and in combination with pre-Initiation Complex (pre-IC) proteins. Furthermore, we want to understand the influence of S-phase specific kinases on helicase activity. Finally, the replication fork architecture and activities associated with the replication fork will be investigated. Newly identified mechanisms will be verified in vivo using a mutagenesis approach. In addition, we will investigate the structure of the MCM2-7 complex in the context of the pre-IC using electron microscopy. Methods include biochemical and biophysical approaches and structural work in an ongoing collaboration with a Cryo EM lab.

You should be highly motivated, committed and have clear career goals. A strong background in biochemistry and protein purification is required. The candidate would benefit from experience in analysis of protein-DNA interactions. You should have a PhD in Biochemistry, Biology or equivalent experience and at least one first-author publication.
