Job description:
A large proportion of tumor samples harbor activating point mutations inthe genes encoding ras family members. Intensive efforts have been madeto determine the importance of effector pathways such as PI3-kinasesignaling in the process of ras-induced tumor formation. Toquantitatively examine changes in the ras signaling cascade that altercellular decisions and promote tumor development, the task will be thegeneration of time-resolved quantitative data to establish a data-basedmathematical model of PI3-kinase and MAP kinase signaling in primaryhepatocytes in response to hepatocyte growth factor (HGF) stimulation.On the cell population level, changes in the dynamic properties ofpathway activation in healthy wildtype versus transgenic mice expressingan oncogenic ras mutant will be examined by quantitative immunoblotting(Schilling et al., FEBS Journal 2005), quantitative protein arrays (Korfet al., Proteomics 2008) and genome-wide time-resolved expressionprofiling. To uncover oscillatory or switch-like behavior changes, inthe subcellular localization of fluorescently labeled components of theras signaling cascade will be monitored by time-lapse experiments. Theaim of the project is to finally predict strategies to correctalterations that promote the progression of cancer.

We are looking for a highly motivated scientist holding a PhD withexperience in tissue culture, biochemical techniques, animal experimentsand a major interest in computer-based analysis to complement our strong team.
The position is available from January 2009 and limited to 31.12.2011.
