Description

Identification and cloning of patient-specific antibodies to breast cancer antigens.

The project involves the use of targeted cDNA cloning to isolate epitope-binding domains of human antibodies that may react with tumor-derived antigens.

The basic approach uses gene-specific PCR and library synthesis, multiplex clone sequencing and in vitro transcription/translation to produce single chain antibodies.The recombinant proteins are then used to trap tumor-derived antigens from patient tumor samples which are identified using sensitive liquid chromatography-mass spectrometry/mass spectrometry (LC-MS/MS) proteomic analysis.

The project requires molecular biology skills (mRNA isolation, PCR, cloning, multiplex plasmid isolation), genetic and database analysis of sequences for protein coding regions and mutations from germline, basic biochemistry and protein analysis such as protein assays, immunoblot analysis and immunoprecipitation.

The project is a unique opportunity to develop a comprehensive screening and analysis path for the identification of functional anti-cancer antibodies. The applicant will be expected to apply the latest techniques in molecular biology to rapidly screen up to 10 independent human antibody libraries and attempt to develop methodologies to screen in vitro translated antibodies for their binding to specific tumor proteins. Career development opportunities include experience in developing efficient cloning process, database management and data interpretation for publication and patent development.
