The Wellcome Trust Sanger Institute has an integrated programme to exploit genomic approaches to study host-pathogen interactions and the analysis of microbial populations. The Microbial pathogenesis team, under the leadership of Professor Gordon Dougan, is focusing on the genetic analysis of the interactions between bacteria and their hosts to shed light on how humans and other animals respond to infection. This information will be exploited towards developing methods of controlling infection, including vaccines and antibiotics. The team is also investigating the relationships between the bacteria that cause typhoid, including Salmonella Typhi, in order to explain their evolution, antibiotic resistance and transmission within the human population. Finally the team is involved in a number of major collaborations worldwide investigating methods for developing vaccines against typhoid and other diseases. As well as focusing on Prof Dougan’s research interests, the Microbial Pathogenesis team provides significant support to other Sanger Institute teams including the Pathogen Genomics group and the Mouse Genetics Programme.

We are now in a position to recruit a candidate for their first Postdoctoral Fellowship. The research project will be the genetic analysis of pathogenic Pseudomonas, characterisation of antibiotic resistance in sequenced organisms and high-throughput trancriptomics of pathogen phenotypes. These are challenging research areas that require an individual with a full understanding of the complex regulation of swarming and surfing motilities in Pseudomona and experience in bacterial genomics and systems biology including working with bacterial complex adaptations, screening large mutant libraries for genes associated with these adaptations, experience with high throughput transcriptomic methods (microarrays and RNA-sequencing), and the ability to investigate bacterial virulence properties including motility, cellular cytotoxicity, biofilm formation as well as determine antibiotic resistance mechanisms.

Essential Skills
• Previous research and laboratory experience of working with Pseudomonad particularly P. aeruginosa
• Experience of routine microbiological procedures including; microscopy, nucleic acid isolation, cDNA synthesis, PCR, primer design, restriction digest, transformation of plasmid/genomic DNA into bacteria via conjugation, electroporation, chemical transformation, complementation studies
• Previous experience in working with type IV pilus-dependent DNA bacteriophage: bacteriophage stocks preparation, plaque formation assays
• Previous experience in setting up and managing screening P. aeruginosa transposon mutant libraries
• Experience in the use of monoclonal antibodies and antisera: ELISA
• Understanding of the motility assays : swimming, swarming, twitching, surfing in Pseudomonads.
• Understanding of static biofilm arrays
• Knowledge of antibiotic susceptibility assays: minimum inhibitory concentration (MIC), kill curves
• Knowledge of Congo red, rhamnolipid, elastase, pyoverdine, pyocyanin assays
• Work knowledge of template preparation for DNA sequencing

Other information
Postdoctoral Fellows are typically in their first or second postdoctoral position as part of a period of early career research training. Successful applicant(s) who have submitted their PhD thesis and are awaiting their PhD award to be confirmed will be placed on a transitional pay point, currently £26,364. On confirmation applicants will be moved on to the pay scale above.

This is a 3 year fixed-term contract
