“Developing novel BAC transgenic mice of C9ORF72-linked motoneuron disease to study RNA cellular pathways”

Applications are invited for a Postdoctoral Research Assistant to join the Laboratory of Molecular Neurodegeneration, a multidisciplinary laboratory focused on elucidating the molecular mechanisms of neurodegenerative diseases.

The project will investigate the function and dysfunction of the newly discovered gene C9ORF72 causing motorneuron disease (MND ) and frontotemporal lobar degeneration (FTLD ). First, we will develop a transgenic mouse model carrying a novel C9ORF72 bacterial artificial chromosome (BAC ) expression construct to visualize alterations in RNA dynamics and will characterize early-stage molecular, histological and behavioral changes. Second, we will model in vitro the development of molecular pathology in human motoneurons derived from induced pluripotent stem cells (iPS) carrying wild-type or mutant variants of the human C9ORF72 gene and by genome editing using zinc finger nucleases in iPS-derived human motorneurons.

The project will be supervised by Dr Javier Alegre-Abarrategui in Dr Richard Wade-Martins’ Molecular Neurodegeneration Laboratory in collaboration with Prof Kevin Talbot. Together, our laboratories integrate studies of neuropathology, genetics and cell biology to study common molecular mechanisms of neurodegeneration.

Applicants must have a PhD/DPhil or equivalent in a relevant area of research and experience in molecular and cellular neuroscience. The appointment is funded by Motor Neurone Disease Association and is offered for up to 36 months, to start as soon as possible.
